Role Description

An opportunity is available for a highly motivated expert in advanced mass spectrometry of HLA-associated peptidomes to join a multidisciplinary group dedicated to understanding and predicting the immunogenicity of protein-based therapeutics. The Principal Scientist will establish the utility of a dendritic cell mass spectrometry (DC-MS) assay to detect peptides from therapeutic proteins that are processed and presented on MHC (major histocompatibility complex) class II for helper CD4 T cell recognition. With prior demonstrated expertise in the application of the DC-MS assay in the field of immunogenicity of protein therapeutics, the successful candidate will implement relevant bioinformatics tools that allow data mining for confident identification of peptide presentation. Alongside the application of the tool to our protein therapeutics portfolio and DC-MS data integration into portfolio decision making, the candidate in this position will work to further refine the technique. The Principal Scientist will join at the interface of predictive immunogenicity sciences and leading protein mass spectrometry analytics to become a key contributor to our efforts to rationally design low immunogenicity biomolecules.


Responsible for the set-up, advancement and application of the dendritic cells mass spectrometry (DC-MS) assay with particular focus on MHCII immunoaffinity procedures, high resolution LC-MS/MS analysis and bioinformatics.Collaborate with Immunogenicity Sciences experts to ensure robust decision making with DC-MS data.As a strong collaborator develop deep partnerships with Immunogenicity Scientists working on cell preparation or assessing immunogenicity project risks as well as other LCMS team members.Participate and lead in technology development and implementation by exploring advancement to the DC-MS workflow.Participate in all phases of mass spectrometry laboratory operations including technology evaluations, the development of sample preparation and separation methods, data analysis, interpretation, reporting and follow up discussions with the project teams on utilizing the data in appropriate decision making.Ensure all tasks and responsibilities are carried out according to scientific and ethical standards e.g. responsible for eWorkbook record keeping.Publications in peer-reviewed scientific journals and external / internal presentations are expected.


Qualified applicants hold a Ph.D. with 5 years of post-doctoral experience including experience in industry, ideally in the application of DC-MS to assess immunogenicity risks of protein therapeutics.Expertise in development and implementation of mass spectrometry based immunopeptidomics methods.Expertise in mass spectrometry based proteomics.Experience and knowledge in operation of QExactive mass spectrometers and associated control and data analysis software systems.Knowledge and application of complex sample preparation methodologies as applied to quantitative protein mass spectrometry workflows techniques including immuno-analytical techniques.Good understanding of protein biochemistry.PHYSICAL/MENTAL REQUIREMENTSAs appropriate for lab based role.NON-STANDARD WORK SCHEDULE, TRAVEL OR ENVIRONMENT REQUIREMENTS Lab based position requiring to perform all typical laboratory related activitiesBased in Andover*Occasional travel to Cambridge (Kendal Square) is required, depending on project needs
